IgM; half maximal binding to coated t-PA at 650 ng/ml (high affinity); reaction with free t-PA and t-PA-PAI-1 complexes; no cross-reaction with urokinase. Directed against an epitope on the light chain of t-PA away from the active site. It stimulates plasminogen activation in the presence of CNBr fragments of fibrinogen and of plasminogen activation in the absence of CNBr fragments of fibrinogen. It can be used in an ELISA-system as catching antibody. Recommended coating concentration 10 µg/ml.
Binds to free t-PA and t-PA-PAI complexes. Reaction with an epitope expressed on the light chain of t-PA different from the active site. No cross reaction with urokinase. Stimulation of plasminogen activation in the presence and in the absence of CNBr fragments of fibrinogen. Half maximal binding to coated t-PA at 650 ng/ml (high affinity).
Hybridization of NSO cells with spleen cells from a BALB/c mouse immunized with purified melanoma-type t-PA.
IgM
The antibody is purified from murine ascites by multiple precipitations with 1% boric acid.
The antibody is lyophilized from a 1 mg/ml solution in isotonic phosphate buffered saline, pH 7.4, containing 0.02% sodium azide and 20 mg/ml mannitol. It is supplied in vials of 500 µg and should be reconstituted with 0.5 mL distilled water.
For extensive dilutions a protein containing solution should be used (e.g. 1% bovine serum albumin in PBS).
Lyophilized antibody should be stored at 4°C. Reconstituted antibody should be aliquoted and stored at -20°C or lower. Avoid repeated freeze-thaw cycles.
