IgG1; binds to single chain urokinase, two-chain urokinase, and low molecular weight urokinase. Partially inhibits the functional activity of two-chain urokinase at appropriate concentrations. Applications include immunoblotting, ELISA tests and enzyme inhibition.

Derivation

Hybridization of NSO cells with spleen cells from a BALB/c mouse immunized with recombinant human single chain urokinase.

Purification

The antibody is purified from murine ascites by affinity chromatography on immobilized recombinant Protein A. The purity of the product is tested by Laemmli SDS-PAGE.

Inhibition of urokinase

  • 14 scu-PA at concentrations ranging from 0-10 µg/ml was preincubated with 20IU/ml urokinase for 15 minutes at 37°C
  • Urokinase activity was detected with a chromogenic substrate (S-2444)
  • Optical density was read after 15 minutes incubation with the chromogenic substrate at 37°C

Handling and storage

The antibody is lyophilized from a 1 mg/ml solution in isotonic phosphate buffered saline, pH 7.4, containing 0.02% sodium azide and 20 mg/mL mannitol. It is supplied in vials of 500 µg and should be reconstituted with 0.5ml distilled water.

For extensive dilutions a protein containing solution should be used (e.g. 1% bovine serum albumin in PBS).

Lyophilized antibody should be stored at 4°C. Reconstituted antibody shouldbe aliquoted and stored at -20°C or lower. Avoid repeated freeze-thaw cycles.
