IgG2b; half maximal binding to coated PAI-1 at 10 µg/ml; reaction with active PAI-1, latent PAI-1 and t-PA-PAI-1 complexes; no cross-reaction with PAI-2 or with PAI-3; interferes with the functional activity of PAI-1. Applications include immunodepletion of PAI-1 from human plasma in ELISA tests.

Specificity

Binds to active PAI-1, latent PAI-1 and t-PA-PAI-1 complexes. No binding to PAI-2 or PAI-3. Interferes with the functional activity of PAI-1.

Derivation

Hybridization of NSO cells with spleen cells from a BALB/c mouse immunized with purified PAI-1 from the human melanoma cell line MJZJ according to the method of Wagner et al. (1).

Purification

The antibody is purified from murine ascites by affinity chromatography on immobilized recombinant Protein A. The purity of the product is tested by Laemmli SDS-PAGE.

Handling and storage

The antibody is lyophilized from a 1 mg/ml solution in isotonic phosphate buffered saline, pH 7.4, containing 0.02% sodium azide. It is supplied in vials of 500 µg and should be reconstituted with 0.5 ml distilled water. For extensive dilutions a protein containing solution should be used (e.g. 1% bovine serum albumin in PBS). Lyophilized antibody should be stored at 4°C. Reconstituted antibody should be aliquoted and stored at -20°C or lower. Avoid repeated freeze-thaw cycles.
