Protein C is activated by a specific activator and measured by its amydolytic activity on a specific chromogenic substrate.

Protein C is a vitamin K dependent serine protease which, when activated, inhibits coagulation by inactivating the clotting factors V/Va and VIII/VIIIa. Additionally, protein C has been shown to have profibrinolytic activity. Hereditary, heterozygous protein C deficiency has been found to be associated with an increased risk of venous thrombosis and hereditary, homozygous total protein C deficiency has been found in neonates with purpura fulminans.
