IgG1; positive reaction exclusively with Glu-plasminogen, in immunoblotting experiments no reaction with Lys-plasminogen; reaction with Plasmin-Alpha-2-Antiplasmin complexes only when Glu-forms are involved in complex formation. Half maximal binding to coated Glu-plasminogen at 6 ng/ml.

Applications include biochemical and pharmacological studies, differentiation between Glu- and Lys- forms of plasminogen, antibody for an ELISA specific for Glu- Plasminogen.

Specificity

Directed against the preactivation peptide. Positive reaction with Glu- Plasminogen and Glu-Plasmin. No reaction with LysPlasminogen. Reaction with Plasmin-α-2-antiplasmin complexes only when Glu- forms are involved in complex formation (1). Half maximal binding to coated Glu-Plasminogen at 6 ng/ml.

Derivation

Hybridization of NSO cells with spleen cells from a BALB/c mouse immunized with purified human plasminogen.

Purification

The antibody is purified from murine ascites by affinity chromatography on immobilized recombinant Protein A. The purity of the product is tested by Laemmli SDS-PAGE.

Handling and storage

The antibody is lyophilized from a 1 mg/ml solution in isotonic phosphate buffered saline, pH 7.4, containing 0.02% sodium azide and 20 mg/ml mannitol. It is supplied in vials of 500µg and should be reconstituted with 0.5 mL distilled water.

For extensive dilutions a protein containing solution should be used (e.g. 1% bovine serum albumin in PBS).

Lyophilized antibody should be stored at 4°C. Reconstituted antibody should be aliquoted and stored at -20°C or lower. Avoid repeated freeze-thaw cycles.
