of Thyroglobulin (Tg) in human serum

Thyroglobulin is an iodine containing protein with a molecular weight of 660 kDa. It is synthesised in thyrocytes and released into follicular lumen. There Tg plays a major role in the production and storage of thyroid hormones thyroxine (T4) and triiodothyronine (T3). The synthesis and the release of Tg is organ and tissue specific and coordinated by thyrotropin (TSH).
For researching differentiated thyroid carcinoma (dtc), neoplasia, or connatal hypothyroidism, the determination of Tg in human serum is a helpful parameter. Non detectable Tg values in hyperthyroid adults could be an indicator for Thyrotoxicosis factitia. In contrast, high Tg values can be used to research different malfunctions of thyroid gland, for example Graves’ disease, Hashimoto’s thyroiditis and non-toxic diffuse goiter. Note, the level of Tg determination is not correlated with the degree of the malfunction.
Medizym® Tg is an ELISA test and adaptable to all open automated systems. Serum Tg is captured by two different monoclonal antibodies specific for two different epitopes of Tg molecule. One monoclonal antibody is immobilized on the wall microtiter well whereas the other is labelled with horseradish peroxidase (HRP) and acts as conjugate.
50 µl of native human serum is needed and total time of incubation is 1 hour and 15 minutes at 37°C under shaking.
